Therapeutic Efficacy of Malachite Green-Based Photodynamic Therapy in Acute Myeloid Leukemia

Aim: Acute myeloid leukemia (AML) is a disease characterized by relapse and treatment resistance in most patients. Therefore, there need for targeted therapies AML. Photodynamic therapy (PDT) promising alternative the of malignant tumors. Also, PDT has potential to be used individually or complementally leukemia. In this study, it was aimed investigate possible effect malachite green (MG)-based on acute cells. 
 Materials Methods: Cells were incubated with 0.19, 0.39, 0.78,1.56, 3.125, 6.25 µM MG one hour irradiated 46.4 J/cm2 light. The trypan blue test assess viability cells, change mitochondrial activity determined MTT. Morphological features Giemsa staining scanning electron microscopy. Cell cycle Annexin V/PI assays (measuring fluorescence emitted reagents) measured flow cytometry.
 Results: With combination light, HL60 cell found significantly reduced compared control group. SEM results showed that 3.125 μM MG-based induced various morphological changes cells typical apoptosis.
